The results of HPLC analysis of Rhodiola rosea before and after fermentation are shown in the figure on the right.

As can be seen from the figure, the retention time of the Rhodiola rosea standard was about 3.7 min, and the retention time of the tyrosol standard was about 11 min. The absorption peaks appeared at 3.8 min and 11 min in the extract of Rhodiola rosea before fermentation.

In the ethanol extract of Rhodiola rosea after fermentation, the absorption peaks at 3.7 min and 10 min were larger than those of the pre-fermentation control. Therefore, it is proved that microbial fermentation can increase the content of rhodiola rosea and tyrosol in Rhodiola rosea.

Fermentation of Rhodiola rosea was carried out at an inoculum rate of 10 % under optimal fermentation conditions, and the contents of Rhodiola rosea xylitol and tyrosol were increased by 52.83% and 11.54%, respectively, as compared with that of the control, which increased the contents of Rhodiola rosea xylitol and tyrosol dramatically, according to the HPLC.

The antioxidant capacity of Rhodiola rosea yeast fermentation product filtrate with different concentrations of the solution to be tested was examined by DPPH free radical scavenging assay. The filtrate of Rhodiola rosea yeast filtrate was diluted with deionized water into aqueous solutions of 10%, 20%, 30%, 40%, 50% and 60% by mass, respectively, as the test solution.

The IC50 of the DPPH scavenging effect of the filtrate of Rhodiola rosea yeast fermentation product was 34.5%, and the scavenging ability of the filtrate of Rhodiola rosea yeast fermentation product on DPPH radicals was gradually increased with the increase of the concentration of the filtrate of Rhodiola rosea yeast fermentation product.

It shows that the filtrate of Rhodiola rosea yeast fermentation product has strong antioxidant ability, which can scavenge free radicals, promote cell metabolism, and play its role in anti-aging.

A solution of 30% by mass of the filtrate of Rhodiola rosea yeast fermentation product was used as the test solution, and 2.0% by mass of the filtrate was used as the sample control. 2.0% arbutin was used as a control for the tyrosinase inhibition assay.

The inhibition rate of 50% of Rhodiola rosea yeast fermentation product filtrate on tyrosinase was better than that of 2.0% arbutin, which indicated that the Rhodiola rosea yeast fermentation product filtrate had obvious ability to inhibit tyrosinase activity in vitro at this action concentration, and was judged to have the ability of whitening.
